proteo]ytic fragments were determined. Screening a cDNA library of MeJA-treated leayes by o]igonucleotide probes designed from the peptide sequences identified a c]one that collld code for a polypeptide with 154 amino acids. The deduced amino acid sequence of ipomoelin showed an oyerall amino acid identity of 25% with the salt-inducib]e SatT protein of rice. In addition, the C-termina] 70 amino acid sequence of ipomoelin showed about 50% identity with the C-terminal amino acid sequences of seed lectins from Mbraceae. The gelle for ipomoelin was present in a few copies in the genome of sweet potato. The mRNA for ipomoelin was deteeted in leayes and petioles, but not in stems and tuberous roots, of sweet potato plants grown in the field. Mechanical wounding of leaves induced ipomoelin mRNA both locally and systemically, while treatment of leayes with ABA, salt, or a high leyel of sucrose did not induce ipomoeli" mRNA. By contrast, ABA-inducible mRNA for sporamin was not induced by MedA. These results suggest that ipomoelin is inyolved in defensiye reactions of leaves in response to wounding and that JA-mediated wound-induction of ipomoelin occurs independe"tly of ABA.

A major jasmonate-inducible protein of sweet potato, ipomoelin, is an ABA-independent wound-inducible protein.

Treatment of sweet potato plants cultured in vitro with a vapor of methyl jasmonate (MeJA) induced an accumulation in leaves of a large amount of protein with an apparent molecular mass of 18 kDa. This protein, designated ipomoelin, was purified, and the amino acid sequences of proteolytic fragments were determined. Screening a cDNA library of MeJA-treated leaves by oligonucleotide probes desig...

Carbon monoxide regulates the expression of the wound-inducible gene ipomoelin through antioxidation and MAPK phosphorylation in sweet potato

Carbon monoxide (CO), one of the haem oxygenase (HO) products, plays important roles in plant development and stress adaptation. However, the function of CO involved in wounding responses is seldom studied. A wound-inducible gene, ipomoelin (IPO), of sweet potato (Ipomoea batatas cv. Tainung 57) was used as a target to study the regulation of CO in wounding responses. After wounding for 1h, the...

The sweet potato IbMYB1 gene as a potential visible marker for sweet potato intragenic vector system.

MYB transcription factors play important roles in transcriptional regulation of many secondary metabolites including anthocyanins. We cloned the R2R3-MYB type IbMYB1 complementary DNAs from the purple-fleshed sweet potato (Ipomoea batatas L. cv Sinzami) and investigated the expression patterns of IbMYB1 gene with IbMYB1a and IbMYB1b splice variants in leaf and root tissues of various sweet pota...

Involvement of hydrogen peroxide and nitric oxide in expression of the ipomoelin gene from sweet potato.

The IPO (ipomoelin) gene was isolated from sweet potato (Ipomoea batatas cv Tainung 57) and used as a molecular probe to investigate its regulation by hydrogen peroxide (H(2)O(2)) and nitric oxide (NO) after sweet potato was wounded. The expression of the IPO gene was stimulated by H(2)O(2) whether or not the plant was wounded, but its expression after wounding was totally suppressed by the pre...

Antagonistic effects of abscisic acid and jasmonates on salt stress-inducible transcripts in rice roots.

Abscisic acid (ABA) and jasmonates have been implicated in responses to water deficit and wounding. We compared the molecular and physiological effects of jasmonic acid (JA) (< or = 10 microM), ABA, and salt stress in roots of rice. JA markedly induced a cationic peroxidase, two novel 32- and 28-kD proteins, acidic PR-1 and PR-10 pathogenesis-related proteins, and the salt stress-responsive Sal...
